Sugar-thioacetamide backbone in oligodeoxyribonucleosides for specific recognition of nucleic acids.

چکیده
The amide linkage being shorter than the natural phosphate linkage, an additional atom is introduced into oligodeoxyribonucleosides (ODNs) with sugar-thioacetamide backbone that show very good RNA recognition properties.

Backbone modification of nucleic acids: synthesis, structure and therapeutic applications.
Nucleic acids have been extensively modified by replacing the phosphodiester group or the whole sugar phosphodiester with alternative anionic, neutral and cationic structures. Several of these modified oligonucleotides exhibit improved properties including enhanced recognition and binding to RNA, duplex DNA and proteins. This has resulted in the development of new and more potent antisense and ...
متن کاملEvolution of acceptor stem tRNA recognition by class II prolyl-tRNA synthetase
Aminoacyl-tRNA synthetases (AARS) are an essential family of enzymes that catalyze the attachment of amino acids to specific tRNAs during translation. Previously, we showed that base-specific recognition of the tRNA(Pro) acceptor stem is critical for recognition by Escherichia coli prolyl-tRNA synthetase (ProRS), but not for human ProRS. To further delineate species-specific differences in acce...
متن کاملNucleic acid binding proteins affect the subcellular distribution of phosphorothioate antisense oligonucleotides
Antisense oligonucleotides (ASOs) are versatile tools that can regulate multiple steps of RNA biogenesis in cells and living organisms. Significant improvements in delivery, potency, and stability have been achieved through modifications within the oligonucleotide backbone, sugar and heterocycles. However, these modifications can profoundly affect interactions between ASOs and intracellular pro...
متن کاملNew insights into the structure of abasic DNA from molecular dynamics simulations
Abasic (AP) sites constitute a common form of DNA damage, arising from the spontaneous or enzymatic breakage of the N-glycosyl bond and the loss of a nucleotide base. To examine the effects of such damage on DNA structure, especially in the vicinity of the abasic sugar, four 1.5 ns molecular dynamics simulations of double-helical DNA dodecamers with and without a single abasic (tetrahydrofuran,...
متن کاملA new conformation-specific infrared band of A-DNA in films.
A band at 1185 cm-1 occurs in the infrared spectra of nucleic acids in their A-type conformation which has been reproduced in the literature, too. The absorbance of this band is proportional to the fraction of the A form of DNA samples containing a mixture of different forms. The 1185 cm-1 band has been assigned tentatively to a vibration of the sugar-phosphate backbone with a fairly high contr...
